摘要:
建立了一个p53蛋白信号网络,包括p53-Mdm2负反馈回路、p53-Wip1-ATM负反馈回路、p53-PTEN-PIP3-Akt正反馈回路.通过分析数学模型再次确认了p53-Mdm2、p53-Wip1-ATM反馈回路是p53振荡出现的必要条件.利用分岔分析等方法,发现低浓度的PTEN能缩短p53的振荡周期,而高浓度的PTEN能终止p53的振荡行为,说明了PTEN的促凋亡作用.结果对理解p53的动力学行为有一定意义.
Abstract:
A p53 protein signal network is established,including a negative feedback loop of p53-Mdm2,a negative feedback loop of p53-Wip1-ATM,a positive feedback loop of p53-PTEN-PIP3-Akt.By analyzing the mathematical model,it is reconfirmed that the feedback loops of p53-Mdm2 and p53-Wip1-ATM are necessary conditions for the emergence of p53 oscillation.By means of bifurcation analysis and other methods,it is found that low concentration of PTEN could shorten the period of p53 oscillation,while high concentration of PTEN could terminate the oscillation behaviour of p53.All these show the apoptosis effect of PTEN.The results have some significances for understanding the dynamic behavior of p53.
