摘要:
细胞中的DNA受损后,肿瘤抑制蛋白p53表现出丰富的动力学,并在决定细胞命运中起到关键的作用.基于PDCD5调控p53-Mdm2振子模型,增加了ATM感应模块,并探讨了在不同PDCD5表达水平下,ATM二聚体转为单体的速率对p53动力学的影响.通过余维一分岔分析,可以看出在不同速率下,p53表现出丰富的动力学,包括单稳态、双稳态、三稳态、振荡以及振荡与单稳态共存.通过对p53动力学的研究,对理解DNA损伤后的细胞命运起到一定的指导作用.
Abstract:
After DNA in a cell is damaged,tumor suppressor protein p53 exhibits rich dynamics and plays a key role in deciding cell fate.An ATM sensor module is added into the p53-Mdm2 oscillator module regulated by PDCD5 and the influence of conversion rate from a ATM dimer to a monomer is discussed on the p53 dynamics at different PDCD5 levels.By codimension one bifurcation analysis of p53 level with respect to conversion rate from the ATM dimer to the monomer at given PDCD5 level,the results show that p53 exhibits rich dynamics,including a stable steady state,two stable steady states,three stable steady states,oscillation and the bistability with a stable steady state and oscillation.The research on studying p53 dynamics will play a guiding role in understanding cell fate after DNA damage.
